علماء يفكون شفرة «كوفيد -19» داخل الرئة.. كيف يتكاثر كورونا ويدمر الخلايا؟

توصل علماء أمريكيون إلى فك الشفرة التي تنظم التفاعل بين فيروس كورونا والرئة، حيث أوضحت الدراسة الجديدة التي أعدّوها في هذا الشأن كيف «يختطف» الفيروس خلايا الرئة البشرية ويدمرها.

وقال تقرير نشره موقع فرست بوست الأمريكي اليوم إن العلماء تمكّنوا من فك شفرة سلسلة الاستجابات الجزيئية داخل خلايا الرئة البشرية، عند الإصابة بعدوى فيروس كورونا المستجد، معتبرًا ذلك تقدما قد يساعد في التعرف على أفضل الأدوية التي تم التصديق عليها بعد التجارب السريرية، والتي يمكن استخدامها في العلاج من الإصابة الفيروسية.

وقامت الدراسة التي نشرت في مجلة «موليكيولار سيل» الطبية، بتقييم خلايا الحويصلات الهوائية من خلال استخدام تكنولوجيا المسح الطيفي الدقيقة، والتي تستطيع تحديد صفات الجزئيات الموجودة داخل العينات التي تخضع للفحص.

وتمكن العلماء بكلية الطب بجامعة بوسطن الأمريكية من التعرف على بروتينات ومسارات الجزئيات في الخلايا الرئوية والتي تتغير مستوياتها بسبب الإصابة بفيروس كورونا، حيث يعتقدون أن النتائج التي توصّلوا إليها تقدم أفكارا عميقة فيما يتعلق بطبيعة المرض والأهداف العلاجية لمنع الإصابة بكوفيد-19.

وتوصلوا إلى أن هناك نوعا من تعديلات البروتين والمعروف بعملية «الفسفرة»، حيث يصبح هذا البروتين شاردا في الخلايا الرئوية المصابة، مشيرين في الدراسة إلى أن عملية فسفرة البروتين تلعب دورا كبيرا في تنظيم وظيفته داخل خلايا الكائن الحي .

والفسفرة أو الفسفتة في الكيمياء الحيوية هي عملية يتم فيها إضافة مجموعة فوسفات إلى جزيء بروتين، أو هو إدخال مجموعة فوسفات إلى جزيء عضوي بشكل عام.

ولاحظت الدراسة أن وفرة البروتين وعملية فسفرة البروتين هما عمليتان تخضعان لمستوى عال من السيطرة عندما تكون الخلايا «صحية»

غير أن العلماء اكتشفوا أن فيروس كورونا يتسبب في حدوث «فوضى» في خلايا الرئة، ليتسبب بذلك في تغييرات غيرعادية في كميات البروتين وتكرار عملية «الفسفرة» داخل هذه الخلايا، مؤكدين أن تلك التغييرات تساعد الفيروس على التكاثر، ومن ثم يدمر الخلايا في نهاية المطاف، ويؤدي إلى انتشار الإصابة على نطاق واسع داخل الرئة.

وأكد الباحثون أن فيروس كورونا يبدأ بسرعة، بعد الإصابة، في استغلال الموارد التي تغذي نواة الخلية الضرورية لنموها ووظيفتها بشكل طبيعي.

ونبه الدكتور بكلية طب بوسطن أندرو إيميلي، والذي شارك في الدراسة إلى أن الفيروس يستخدم هذه الموارد للانتشار بينما يتجنب الهجوم من جهاز المناعة في الجسم، وبهذه الطريقة، تتكون الخلايا الجديدة والتي تتطرد الخلايا المنهكة والخلايا الرئوية التي تم هدمها بشكل وحشي، لتتركها تدمر نفسها ،مضيفا أن هذه الفيروسات الجديدة تنشر العدوى في الخلايا الأخرى، حيث تتكرر نفس الدورة .

وفحص العلماء في الدراسة خلايا الحويصلات الهوائية لفترات تبدأ من ساعة وحتى 24 ساعة بعد الإصابة بفيروس كورونا، وذلك حتى يتسنى فهم التغيرات التي تحدث في هذه الخلايا بشكل مباشر، أي بعد ساعة أو ثلاث أو ست ساعات من الإصابة، ومن ثم التعرف على التغيرات التي تحدث في وقت لاحق .

وأكد الدكتور إيلك مولبيرجير، الذي شارك في الدراسة أن المعلومات التي توصلوا إليها توضح أن فيروس كورونا يتسبب في عدد هائل من التغيرات داخل الخلية المصابة تبدأ بعد ساعة واحدة من الإصابة وتضع الأساس لما أسماه بـ«اختطاف كامل» لخلايا الرئة المضيفة.
